Know the benefits of the formats of a mini spin column

A mini spin column plays a very important role during the chromatographic performance. When these columns are well packed, they will not only offer reliable performance in the entire design process. They will also be capable of delivering a constant product recovery as well as separation from pollutants. Additionally, a good design of these columns will deliver augmented resolution between peaks, make the process of packing of multiple types of resins easy at diverse bed heights, and offer scalability from process growth to full level manufacturing.

A mini spin column is appropriate for isolating the poly A+ mRNA from a maximum of 1 mg total RNA, 100 mg tissue, or 2 x 107 cells. These spin columns are available in mRNA as well as in mRNA mini and micro kits. Fast viewing of chromatographic conditions is essential to spot the best cleansing conditions. Investigation of conditions can be accomplished on laboratory scale automated robotic systems or chromatography systems. PRC Robo Columns, as well as Pre-Packed Columns for viewing are strong tools for rapid purification development. Both products are available in a pre-packed condition with a mixed mode, ion exchange, as well as with affinity chromatographic materials.

Both the PRC Pre-Packed, as well as the Robo mini spin column will aid you greatly in finding the accurate solution for your chromatography process. Robo micros spin columns are being delivered in strips of 8 pre-packed components for screening purposes. They are available in two different formats, such as 200 µL units and 600 µL units.

While the 200 µL units are available with a bed height of 10 mm, the 600 µL units are available with a bed height of 30 mm. Similarly, 200 µL units are suitable for screening cleansing conditions and choosing chromatographic materials. This mini spin column format is suggested for screening sorbent and process conditions.

On the other hand, the 600 µL units are suggested when users need residence times for a longer period, usually for more than 4 minutes, such as for the assessment of dynamic binding volumes. The 600 µL unit spin column format is recommended for optimizing the process of purification. They are specifically designed for fully automatic as well as for parallel chromatographic isolations with automatic liquid handling workstations. Both formats of the mini spin column are being delivered as rows of eight units, with two detachable silicon cover seals for appropriate storage.

Reaping the real benefits of using a mini spin column mainly counts on its format as well as on high-quality packing. This will enable the skill to mimic the entire steps of the process of the chromatography isolation in dynamic mode. Facts that are attained from smaller columns are comparable to those received in bigger columns.

Some of the other benefits of using these spin columns include:

·         The miniaturized format of these columns eases viewing of chromatography sorbents, lessens sample use, as well as puts the time aside.

·         The format enables users to meet the fitting requirements of the DoE or the Design of Experiment, allowing a huge amount of tentative work in a minimum time.

·         The format also allows trouble-free integration with computerization systems for HTPD or High Throughput Process Development.

Above all, both mini spin column formats allow users to test multiple parameters in parallel.

How does the mini spin column help to purify nucleic acids?

Nucleic acids can be purified using spin columns, as well. Purification of these acids, which is based on spin columns, is a solid stage extraction technique to purify them quickly. This method counts on the fact that nucleic acids will attach to the solid stage of silica under definite conditions.

Mini Spin Column has an all-in-one Silica Membrane, which has been extensively used as an economical substitute for Qiaquick, Qiaprep, Qiaamp, RNeasy, DNeasy, PureLink, PureYield, GeneElute, and more. In a separate single-column design, it is used in a variety of applications, ranging from Deoxyribonucleic acid to Ribonucleic acid, from normal PCR purification to the preparation of the next generation sequencing sample, and from plasmid to viral and genomic DNA.
Since the introduction of these spin columns, millions of them served scientists all over the world. Buffs have honored them in peer-evaluated publications with their individual mission-customized buffers for an assortment of applications. They are compatible with an extensive variety of buffers that carry different brand names.

The procedure involves different methods and stages to purify nucleic acids. The different stages include lyse, connect, clean, and elute. This means that lysis of cells, joining of nucleic acids to silica gel membrane, cleaning the nucleic acids connected to the silica gel membrane, and removal of the nucleic acids.

During the lyse stage, the cells of a sample are opened with a lysis method. This will break the cell membrane as well as the nucleus to discharge the nucleic acid.

During the binding stage, a buffer solution is added to the sample, together with isopropanol or ethanol to form the binding solution, which will then be transferred to the spin column. Then, the column is put in a centrifuge, which will force the binding solution through the inside silica gel membrane of the mini spin column. If the salt concentration and pH of the binding solution are most favorable, the nucleic acid will connect to the silica gel membrane while the solution traverses.

To clean, the flow-through is detached and a clean buffer is attached to the column. Again, the column is put in a centrifuge, forcing the clean buffer through the membrane. This will remove any residual contaminants from the membrane, leaving only the silica gel connected nucleic acid
During the elution stage, the wash buffer is taken away and water or an elution buffer is added to the spin column. Once again, the column is put in a centrifuge, forcing the removal buffer through the membrane, making the buffer to take the nucleic acid away from the membrane and it is collected from the column bottom.

Maxi spin column, which also includes a Silica membrane, is renowned for its extremely high binding capacity. This makes these spin columns run experiments with higher amounts of Deoxyribonucleic acid to acquire the curve completely to saturate. This binding will be higher than that of other columns.

Some of the other beneficial features of these spin columns include:
·         They can be used to extract both Deoxyribonucleic acids as well as Ribonucleic acids,
·         As the spin column comes with enough amounts of layers, it will offer a guaranteed result.
·         These columns are highly qualified for the purification of DNAs and RNAs.
·         The spin column is capable of dealing with maxi 50-ml samples.
Above all, the unique even-bottom-basket design of the maxi spin column ensures no liquid remains.

All you need to know about mini and maxi spin columns

A mini spin column is a popular type of DNA column that has been extensively used as an economical substitute for plasmid DNA by major manufacturers. These columns are also the perfect substitutes for  viral as well as for a range of genomic DNA Kits including :

·         Qiagen, such as Qiaprep, QiaAmp, Qiaquick, and DNeasy
·         Sigma, such as GeneElute
·         Promega, such as PureYield DNA
·         Invitrogen, such as PureLink  DNA
Many manufacturers of these columns offer solution recipes for DNA gel extraction, DNA mini-prep, and PCR clean up for their customers who want to build their own solutions.

Some of the different types of Mini Spin Column Kits include:

DNA Gel Extraction Kits: These kits are designed with Membrane technology in the spin column set-up. They are suitable for extracting DNA from the agarose gel. They are the most useful columns, as well, if the starting DNA is more than 10 micro g
DNA Isolation Kits: The same Membrane technology is also used to design these kits. They can be effectively used in phage, plasmid, and genomic DNA separation.

DNA Clean-up or Purification Kits: As the name suggests, these kits are intended for purifying and concentrating DNA. Users can exploit the Mini Spin column if the starting material is not restricted.
PCR Clean-up or Purification Kits: These kits are used in the same fashion as that of the DNA Clean-up or Purification Kits. However, they are designed to clean and purify PCR.

A Maxi Spin Column is available in different volumes according to the needs of users. It is broadly categorized into two types, such as RNA maxi spin columns and DNA Maxi Spin columns.  Both types of maxi spin columns are capable of providing users with an easy, fast, and cost-efficient way for isolating RNA and DNA from different samples in a large level. They can be also be used for diverse applications by making use of different protocols and buffers.

Similar to a Mini and Tini spin columns, the maxi spin columns hold a silica-based membrane. It is well suited with all the buffers, which are used for Tini and Mini spin columns.  Users can exploit these columns with both centrifuges as well as with vacuum manifolds.   They can connect the Maxi filter column to a Maxi spin column for clarifying cell lysate by making use of vacuum manifold.

Typical maxi spin columns that are available with the binding Capacity of 1000ug are mostly used in the process of nucleic acid purification of size less than 300kb from genomic and plasmid DNA lysed cultures. Each of these columns is capable of processing cell cultures with varying capacities, ranging from 20 ml to 50 ml.

It should be noted that a purification assistant using the silica membrane in the form of a column is used in the process of RNA or DNA purification or extraction of nucleic acids of the highest quality. A maxi spin column is compatible with store-bought buffers as well as with in-house ones. These columns are available in 20 and 5 number packages.
